Officers said when they arrived in the area and spoke to a witness who called 911, they found a vehicle in the St. Marys River just south of the bridge across from Swinney Park.
Police said the vehicle had one man inside who was determined to be in life-threatening condition.
At this time, it appears the vehicle was traveling westbound and for some unknown reason, left the roadway and entered the river.
About an hour later detectives said the man was pronounced dead at the hospital.
The identity, as well as the cause and manner of death, will be released by the Allen County Coroner's Office at a later time.
There were no other vehicles involved and no other injuries.
Investigators said they are trying to piece together what happened and will take measurements and interview any witnesses to determine why the vehicle left the roadway.
Police said eastbound traffic on Jefferson was shut down for several hours.
Motorists are asked to avoid the area.
Westbound traffic was shut down until about 7:30 a.m.
